Operating Parameters Optimization for the Production of Liposomes Loaded with Antibodies Using a Supercritical Fluid-Assisted Process

Encapsulation of antibodies represents a significant advance to protect and deliver these therapeutics in controlled manner, increasing the stability requested cover temporal gap between particle production their administration. Furthermore, using encapsulation, extracellular, cell surface, intracellular targets can be reached. This work examines feasibility encapsulating mouse IgG isotype control within phosphatidylcholine-based liposomes supercritical fluid-based process called SuperLip (Supercritical-assisted Liposome formation). allows continuous both nano- micrometric with high encapsulation efficiency working under mild operative conditions. The effect some parameters has been studied on liposome mean diameter, size distribution, antibody entrapment efficiency, comparing data those collected obtained by thin-layer hydration technique. In particular, water flow rate loading were studied. Antibody-loaded diameters range 205 501 nm have fluid-assisted process. High efficiencies up 94% calculated.
